IV. Cycle Sequencing

Cycle Sequencing Mix (Applied Biosystems Big Dye Termination Kit)

Make 10 µl in a tube suitable for your PCR heating block.

Make fresh and keep on ice. Do not refreeze.

  1. Add 1 µl of primer (5'mt Control Region [6 pm/µl])
  2. Add 1 µl of Big Dye Termination Mix
  3. Add 4.5 µl of distilled water
  4. Add 1.5 µl of 5x Sequencing buffer
  5. Pipette up and down to mix.
  6. Add 2 µl of PCR product for template DNA
